Native peoples lived in the area at least as early as 11,000 B.C., and when Europeans arrived it was home to the Illini, Osage and Missouri. According to a later collection of legends, the French Canadian Louis Blanchette met Bernard Guillet, an honorary chief of a Dakota band, at the site in 1765, and Guillet told him he had named it Les Petites Cotes, "the little hills," for the hillsides. Blanchette founded the settlement around 1769 under the Spanish governor of Upper Louisiana, which France had ceded to Spain after the Seven Years' War, and served as its civil and military leader until his death in 1793. The settlers were mainly French Canadians and Native Americans, and it is the third-oldest city in Missouri.
The first church, built in 1791, was Catholic and dedicated to St. Charles Borromeo under his Spanish name, San Carlos Borromeo, and the town became known as San Carlos del Misuri. The Spanish lieutenant governor Carlos de Hault de Lassus appointed Daniel Boone commandant of the Femme Osage District, a post he held until the United States took control in 1804 after the Louisiana Purchase, when San Carlos became St. Charles. William Clark arrived on May 16, 1804, with 40 men and three boats to prepare for the expedition west while waiting for Meriwether Lewis to come from St. Louis. The Boone's Lick Road, considered to begin in St. Charles, became the main overland route for settlement of central and western Missouri, ending at Franklin, where the Santa Fe Trail began.
